Lysosomal pathway of the TRAIL-induced apoptosis in breast carcinoma cells-the role of the lysosomal protease cathepsin D

| Popis | In this study, we investigated the role of lysosomal aspartyl protease cathepsin D (CTSD( in the TRAIL-induced apoptosis. We showed that overexpression of CTSD sensitizes MDA-MB-231 breast carcinoma cells to the TRAIL-induced apoptosis in enzymatic activity-dependent manner. The increased frequency fo apoptosis observed in the CTSD-overexpressing cells corresponded to caspase activation and cleavage of the Bid protein, the pro-apoptotic BCL-2 family member. We also showed that the CTSD-mediated pro-apoptotic effect can be partially reversed by the caspase 8 and aspartyl protease inhibitors. |
